In normal whole blood samples, the erythrocyte concentration is a thousand times higher than that of leucocytes. Erythrocyte contamination is therefore often a problem when examining or analysing leukocytes.
Individual lysis of red blood cells is possible with our special erythrocyte lysis buffer. The erythrocyte lysis buffer has been optimised for the lysis of red blood cells from EDTA/citrate/heparin-treated whole blood, buffy coats (PBMCs, leukocyte films) and bone marrow and shows only minimal effects on lymphocytes when used as directed.
The erythrocyte lysis buffer is 1x concentrated (ready-to-use) and contains ammonium chloride. Ammonium chloride lyses red blood cells. Cells with nuclei are not effectively lysed with ammonium chloride.
Good to know
The erythrocyte lysis buffer is mainly used for the lysis of human erythrocytes in single cell suspensions from peripheral blood or haematopoietic tissue as well as spleen from mice. The removal of red blood cells is also often a necessary step prior to flow cytometric analyses, immunophenotyping, immunofluorescence staining, cell culture, cytospin, cytogenetics and cell sorting.
