HEK-293 Medium A is a serum-free, ready-to-use medium optimized for the cultivation of adherent HEK-293 cells. The medium is based on DMEM and has been specially enriched with L-glutamine, cholesterol, albumin, vitamins, hormones, growth factors, and additional trace elements (total content of proteins and animal components < 0.2% w/v).

Adaption von HEK-293-Zellen

Ein Wechsel von serumhaltigem Medium auf unsere HEK-29 Medien ist häufig ohne eine besondere Adaption möglich. Für Zellklone, die ein einfaches Umsetzen in HEK-Medium serumfrei nicht vertragen, empfehlen wir die Zellen in HEK-Medium serumfrei mit Serumzusatz zu kultivieren und den Serumanteil allmählich zu reduzieren. Unterstützt wird dieses Verfahren durch höhere Zelldichten bei der Aussaat.

Für eine erfolgreiche Überführung der Zellen in eine serumfreie Kultur spielt der Vitalitätszustand eine entscheidende Rolle. Daher müssen die Zellen in der logarithmischen Wachstumsphase entnommen werden. Nach unseren Erfahrungen gelingt die Anzucht aus der logarithmischen Phase mit deutlich höheren Erfolgsaussichten.

Bei der Umsetzung der Zellen muss sichergestellt werden, dass nach einer eventuellen Ablösung der Zellen mit Trypsin das Enzym durch vollständiges Auswaschen entfernt bzw. durch einen Trypsin-Inhibitor gehemmt wird, da der im Serum vorhandene Neutralisationseffekt wegfällt.
