Pfu polymerase is a thermostable DNA polymerase. The abbreviation Pfu stands for Pyrococcus furiosus, a thermophilic archaebacterium from which this polymerase was isolated.

The Pfu polymerase ensures through its "read ahead function" or also "proof-reading activity" that fewer faulty PCR products are synthesised. In molecular biology, Pfu polymerase is mainly used in those PCR reactions that have to deliver sequence-exact DNA amplificates.

    The accuracy of DNA synthesis is increased 12-fold compared to Taq DNA polymerase, the error rate is 0.2 x 10-5.
    The PCR products generated with Pfu DNA Polymerase have smooth ends ("blunt ends") and can be used directly in ligation reactions.
    The extension rate of Pfu DNA polymerase is 0.5 kb/min, therefore 1 - 2 min extension time/kb fragment length is recommended.
