Keratinocyte medium - with supplements, 500 ml

Our Keratinocyte Medium is a basal medium for the serum-free culture of human keratinocytes. The formulation of our Keratinocyte Medium is based on the MCDB 153 medium. It selectively supports the growth of human keratinocytes while suppressing the overgrowth of fibroblasts. The basal medium must be mixed with the supplied supplement kit before use.

The supplement kit contains:

• Ethanolamine
• Epidermal growth factor (EGF)
• Insulin
• Hydrocortisone
• Pituitary extract (BPE)
• Phosphoethanolamine

Here’s how to easily supplement your medium:

• Thaw the supplements at room temperature.
• Add the supplements to the basal medium (ensure sterility).
• Trypsinize the keratinocytes, wash them with PBS, and neutralize any remaining trypsin with trypsin inhibitor (10 mg/ml).
• Perform another wash step (PBS) and then determine the cell count. For subculturing, adjust the cell density to 3,000 to 5,000 cells/mL.
  (Initial culture: 1–5 × 10⁶ cells/cm²). Incubate keratinocytes in serum-free supplemented keratinocyte medium in an incubator.
• Alternatively, coat culture dishes beforehand with adhesion factors (collagen, fibronectin, or natural or synthetic fragments of these factors).
