SCRIPTUM-55 Reverse Transcriptase (RT) (200 U/µl) is an M-MuLV reverse transcriptase that rapidly and specifically generates cDNA. The genetically modified enzyme has a high thermostability up to 55°C and is well suited for the reverse transcription of RNAs with complex secondary structures. Our SCRIPTUM-55 RT has a very high RNA affinity and is ideal for assays with low-concentration target RNA or low copy genes. Due to its high purity, SCRIPTUM-55 RT is also ideal for the detection of pathogens. For best results, our SCRIPTUM-55 Reverse Transcriptase is supplied with a specifically optimised, 5x concentrated reaction buffer.

Good to know

Single-stranded nucleic acids often form intramolecular double strands through hydrogen bonds between complementary sections. This results in secondary structures with so-called ‘stems’ (paired double-stranded sections) and ‘loops’ (double-stranded sections that include single-stranded sections). RNA secondary structures can take various forms such as hairpin loops, interior loops or multi-loops.

To analyse an unstable RNA target structure, it must first be transcribed into a stable cDNA by a reverse transcriptase. However, complex RNA secondary structures make it impossible for the reverse transcriptase to attach to the RNA or to read the information. In order to resolve complex RNA secondary structures, the assay temperature must be increased. However, human enzymes begin to denature at 42°C. Our SCRIPTUM-55 RT has been genetically modified and has a thermostability of up to 55°C, which means that reverse transcription of complex RNA secondary structures is not a problem.
